Hydroxylysine glycosides in human urine.

چکیده
Hyd roxylysyl-galactosyl-glucose (HGG) and hyd roxylysyl-galactose (HG) were isolated from normal human urine and shown to be identical with hydroxylysine glycosides (OHLG’s) whose structure had been elucidated in other laboratories. We devised a procedure for separating the OHLG’s from many other urinary constituents by preliminary fractionation on a column of Sephadex. The OHLG’s in the fractions so obtained were then isolated, uncontaminated with other ninhydrin-positive components, in a single pass through an analytical cation-exchange column equipped for splitting the effluent stream. In addition, a procedure was devised for determining as little as 10’ mol of the OHLG’s in 100Mlof filtrate (corresponding to 83.3 ,d of urine) on standard ion-exchange chromatographic amino acid analyzers. In normal adult urine, 1.5,moI of each glycoside was present per 100 mg of creatinine; the molar ratio of HHG to HG was about 1.1 to 1.2. Each was present in plasma in about 400to 500-fold smaller concentration than in urine. After surgical fusion of the spine, excretion of OHLG’s increased and the ratio of HGG to HG decreased, changes we interpreted as indicating a more rapid bone-collagen turnover consequent to surgical damage.

Urinary excretion of hydroxylysine and its glycosides as an index of collagen degradation.
Urimary excretion of hydroxyprolin (Hyp) is one index of total collagen degradation, from all sources. Since some of the Hyp released from collagen may be further metabolized before it is excreted, other markers are necessary to measure collagen breakdown. Excretion of the glycosides of hydroxylysine (Hyl), glucosyl galactosyl hydroxylysine (Hy1[Gl)cGa1]), and galactosyl hydroxylysine (Hyl[Ga)]...
متن کاملBiochemical properties of human glomerular basement membrane in normal and diabetic kidneys.
To determine the presence of any significant structural abnormalities in the glomerular basement membrane (GBM) of diabetic individuals, GBM from normal and diabetic human kidneys were isolated and analyzed chemically and structurally. The amino acid composition of the normal GBM revealed the presence of significant amounts of hydroxyproline, hydroxylysine, glycine, and carbohydrate suggesting ...
متن کاملDetermination of hydroxylysine in urine.
A new method for colorimetric determination of urinary hydroxylysine is described. Approximately one-hundredth of human urine collected for 24 hr was diluted to 25 ml and titrated to pH 2.0 with 2 hcl, and then subjected to column chromatography on Dowex 50 X 4 (H+ form). Amino acids were eluted from the column with 1.5 N NH4OH. Hydroxylysine in the elute with 1.5 N NH4OH was separated from th...
متن کاملCollagen markers deoxypyridinoline and hydroxylysine glycosides: pediatric reference data and use for growth prediction in growth hormone-deficient children.
BACKGROUND In children and adolescents, markers of bone and collagen metabolism reflect the dynamics of skeletal growth and development. The aim of this study was to assess the relationship of the urinary collagen markers deoxypyridinoline (DPD) and hydroxylysine (Hyl) and its glycosides [galactosyl-Hyl (Gal-Hyl) and glucosyl-Gal-Hyl] with growth. METHODS Urine samples from 240 apparently hea...
متن کاملThe isolation, and amino acid and carbohydrate composition, of polymeric collagens prepared from various human tissues.
1. Insoluble polymeric collagens from various human tissues were prepared by the EDTA method. Almost all of the collagen from simple soft tissues such as dermis, tendon, submucosa, sclera and cornea could be extracted, whereas the more complex tissues such as intercostal cartilage and intervertebral disc yielded only small amounts of collagen. Amino acid and carbohydrate analysis indicated that...
